RNA–protein complexes (RNPs), formed when proteins recognize the sequence and structure of RNA, regulate gene expression and cell fate decisions and play a central role in the molecular mechanisms inside cells. The Saito Lab takes on the development of new technologies for understanding and controlling living systems, through design biology and experimental evolution centered on RNA and RNPs. The lab provides research supervision to master's and doctoral students in the Graduate School of Medicine, Kyoto University, helping them acquire methods in RNA synthetic biology, molecular biology, stem cell biology, and bioengineering.
Yoko Hamazaki, Professor

The immune system not only protects the body from infection, but is also deeply involved in the rejection of transplanted organs and in the onset of age-related diseases such as lifestyle-related diseases and cancer. The Hamazaki Lab mainly studies T cells, which act as the command center of the immune system, and the thymus, the organ that produces them. The lab aims to explore the basic principles of immunology and, building on them, develop new ways to control immunity, thereby contributing broadly to medicine and healthcare.

iPS cells can be used to reproduce organ development in a culture dish to make cells for transplantation, and to reproduce disease mechanisms to aid drug development. However, for diseases involving many types of cells, it is difficult to reproduce the disease state with iPS cells alone, and model animals such as mice are also indispensable. The Kawaguchi Lab studies endoderm-derived organs, including the pancreas, carrying out research using iPS cells and research on organ regeneration, aging, and cancer at the same time. This is because organ development and the cell behavior seen in regeneration, aging, and cancer share common features, such as active proliferation and cell migration.
Yasuhiro Takashima, Professor

Stem cells continually produce new cells in the body and replace aged ones, supporting us every day. The Takashima Lab aims to promote health by controlling stem cells. The lab has succeeded in establishing naive human iPS cells, which are closer to a fertilized egg, and found that they can differentiate into cells of all lineages of the gestational period, including the placenta and yolk sac. It has also succeeded in creating stem cell-based embryo models that mimic human embryonic development.
Takuya Yamamoto, Professor

The Yamamoto Lab leverages iPS cell technology to pioneer new fields of life science at the molecular and cellular levels. Focusing mainly on cell fate conversion, including the process by which iPS cells are induced, and on aging of the whole organism, the lab seeks to understand how information is retrieved from genomic DNA and linked to cell functions and properties. It analyzes in an integrated way regulatory mechanisms spanning many levels, such as higher-order chromosome structure, epigenetic regulation, transcription, and translation.

The sequence of the human genome can now be read on a wide scale, but most of the functions encoded in it remain a mystery. Human iPS cells carry the same genome as the donor and can be differentiated into the cell types affected by disease, which makes them a powerful tool for studying genome function. Woltjen Lab specializes in cutting-edge genome editing technologies in iPS cells. Through the three Rs, "Read," "wRite," and "Regulate," the lab aims for complete control of the human genome. Many lab members are international researchers, creating an international and diverse research environment.

Advances in technology are prompting a reexamination of the classical central dogma, in which RNA transcribed from DNA is translated into protein. Takahashi Lab focuses on molecules that fall outside the classical central dogma, such as yet-undiscovered small proteins and functional RNAs. With this new perspective, and using pluripotent stem cells as a model, the lab aims to uncover how cell fate is determined.
Shunsuke Shimobayashi, Associate Professor

Molecules inside cells spontaneously assemble to form organelles that are not enclosed by lipid membranes, and these organelles play important roles throughout the cell. Shimobayashi Lab focuses on the statistical properties of molecules in cells, including phase separation, to uncover the roles of membraneless organelles. In particular, the lab studies the principles of reprogramming, which turns back the time of cells, using approaches that go beyond conventional molecular biology, such as super-resolution imaging, optical control of molecular assembly, and mathematical modeling.

iPS cells are pluripotent stem cells that can differentiate into many cell types. They are made by introducing specific transcription factors into somatic cells to reprogram them. The Nakagawa Lab conducts basic research that aims to reach the essence of cellular plasticity by elucidating the mechanisms of reprogramming. The lab found MYCL, a safer substitute for c-MYC, and has established a method that achieves both efficiency and quality in generating iPS cells.

Proteins are mainly what directly build and carry out the structure and function of cells, yet cell states have until now been discussed mostly on the basis of RNA levels. It is known that RNA abundance and protein abundance are not necessarily proportional. The Iwasaki Lab develops proteome analysis technology with high accuracy and reproducibility, and uses it to uncover the principles that regulate protein abundance, translation, and degradation in pluripotent stem cells. A protein's location within the cell, its post-translational modifications, and how it forms complexes also determine the function of each cell type. iPS cells, which have the same genome yet can differentiate into diverse cell types, are an ideal model for exploring the principles that regulate molecular abundance.
